Short talk description: The seminar will focus on the most important structural and functional aspects of hyaluronic acid. In particular, the functional roles of the biopolymer and its involvement in human pathology including inflammation and tumours will be described. The biotechnological aspects that make this product one of the most widely used polymers in aesthetic and regenerative medicine will also be described. The chemical versatility of polymer will therefore be the starting point for outlining possible new developments of new biotechnological products for the controlled release of drugs and the creation of useful scaffolds for regenerative medicine.

Short cv: Alberto Passi is full professor of biochemistry at the School of Medicine of the University of Insubria, Varese Italy. He serves as Dean of the Medical School and is Director of the Clinical Chemistry Lab of the University Hospital in Varese. His scientific interests have always focused on the biochemistry and biology of the extracellular matrix with particular attention to hyaluronic acid.

Short cv: Liliana Schaefer, MD, is a Professor of Pharmacology at the Institute of Pharmacology and Toxicology, Goethe University in Frankfurt, Germany and Adjunct Research Professor of Pathology, Anatomy and Cell Biology, Thomas Jefferson University, Philadelphia, USA. Dr. Schaefer has made significant contributions to the field of proteoglycans by discovering that two components of the extracellular matrix, Decorin and Biglycan, act as endogenous danger signals. Currently Dr. Schaefer is the President of the Histochemical Society, the Editor-in-Chief of the American Journal of Physiology-Cell Physiology, the member of the FASEB publication committee, and the Chair of the Gordon Research Conference on Proteoglycans 2022.

Short talk description: It is well established that biglycan, a small leucine-rich proteoglycan, acts as an extracellular matrix-derived danger signal in its soluble form. By binding to innate immunity Toll-like receptors (TLR) 2 and 4, biglycan initiates and perpetuates the inflammatory response. Previous work has conveyed that biglycan's role in inflammation extends far beyond its function as a canonical danger signal. Mounting evidence suggests that the selective interactions between biglycan, TLRs, and their adapter proteins critically regulate downstream signaling and disease outcome. Biglycan can act as a high-affinity ligand for TLR coreceptors CD14 and CD44, further providing an additional layer of complexity. We propose a novel concept, that biglycan steers signaling toward inflammation by interacting with CD14, whereas it can trigger autophagy by binding to CD44. Thus, biglycan, and perhaps others soluble proteoglycans, could function as molecular switches which could either propagate the signaling of chronic inflammation or promote the resolution of inflammatory processes.

Short talk description: Heparanase (HPSE) has been defined as a multitasking protein that exhibits a peculiar enzymatic activity towards HS chains but which simultaneously performs other non-enzymatic functions. Through its enzymatic activity, HPSE catalyzes the cutting of the side chains of heparan sulfate (HS) proteoglycans, thus contributing to the remodeling of the extracellular matrix and of the basal membranes. Furthermore, thanks to this activity, HPSE also promotes the release and diffusion of various HS-linked molecules like growth factors, cytokines and enzymes. In addition to being an enzyme, HPSE has been shown to possess the ability to trigger different signaling pathways by interacting with transmembrane proteins. In normal tissue and in physiological conditions, HPSE exhibits only low levels of expression restricted only to keratinocytes, trophoblast, platelets and mast cells and leukocytes. On the contrary, in pathological conditions, such as in tumor progression and metastasis, inflammation and fibrosis, it is overexpressed. With this talk , we intend to provide an update on the current knowledge about the different role of HPSE during tumor progression and fibrosis.

Short cv: Prof. Onisto graduated from the Biological Sciences at the University of Padova in 1985. During 1988-1992 he worked as a Ph.D. candidate in molecular and cellular biology and pathology at University of Padova. In 1992 became Assistant Professor at the Institute of Histology and Embryology (from 2000 at the Dept.of Biomedical Sciences), School of Medicine of the University of Padova and Associate Professor in 2016.  His current research deals with the study of heparanase, the unique and specific endoglycosidase capable of cleaving heparan sulfate (HS) chains. HS cleavage results in remodelling of ECM as well as in regulating the release of many HS-linked molecules such as growth factors, cytokines and enzymes involved in inflammation, wound healing and tumour invasion. Dr. M. Onisto research activity has resulted in 89 publications (83 papers on international journals surveyed by ISI and 6 book chapters)

David Rand is the Erwin H. Schell Professor and Professor of Management Science and Brain and Cognitive Sciences at MIT, an affiliate of the MIT Institute of Data, Systems, and Society, and the director of the Human Cooperation Laboratory and the Applied Cooperation Team. Bridging the fields of cognitive science, behavioral economics, and social psychology, David's research combines behavioral experiments run online and in the field with mathematical and computational models to understand people's attitudes, beliefs, and choices. He focuses on illuminating why people believe and share misinformation and "fake news," understanding political psychology and polarization, and promoting human cooperation. David received his B.A. in Computational Biology from Cornell University in 2004 and his Ph.D. in Systems Biology from Harvard University in 2009, was a post-doctoral researcher in Harvard University's Department of Psychology from 2009 to 2013, and was an Assistant and then Associate Professor (with tenure) of Psychology, Economics, and Management at Yale University prior to joining the faculty at MIT. He has published over 175 papers in peer-reviewed journals including Nature, Science, Proceedings of the National Academy of Science, the American Economic Review, Psychological Science, Management Science, New England Journal of Medicine, and the American Journal of Political Science, and has received widespread attention from print, radio, TV and social media outlets. He has also written popular press articles for outlets including the New York Times, Wired, New Scientist, and the Psychological Observer. He was named to Wired magazine's Smart List 2012 of "50 people who will change the world," chosen as a 2012 Pop!Tech Science Fellow, awarded the 2015 Arthur Greer Memorial Prize for Outstanding Scholarly Research, chosen as fact-checking researcher of the year in 2017 by the Poyner Institute's International Fact-Checking Network, awarded the 2020 FABBS Early Career Impact Award from the Society for Judgment and Decision Making, and selected as a 2021 Best 40-Under-40 Business School Professor by Poets & Quants. Papers he has coauthored have been awarded Best Paper of the Year in Experimental Economics, Social Cognition, and Political Methodology.

 Περίληψη

The spread of misinformation online is a global problem that requires global solutions. To that end, we conducted an experiment in 16 countries across 6 continents (N = 33,480) to investigate predictors of susceptibility to misinformation and interventions to combat misinformation. In every country, participants with a more analytic cognitive style and stronger accuracy-related motivations were better at discerning truth from falsehood; valuing democracy was also associated with greater truth discernment whereas political conservatism was negatively associated with truth discernment in most countries. Subtly prompting people to think about accuracy was broadly effective at improving the veracity of news that people were willing to share, as were minimal digital literacy tips. Finally, crowdsourced accuracy evaluation was able to differentiate true from false headlines with high accuracy in all countries. The consistent patterns we observe suggest that the psychological factors underlying the misinformation challenge are similar across the globe, and that similar solutions may be broadly effective.

Short cv: María dM. Vivanco graduated from the University of the Basque Country, worked at Sandoz (Basel) and carried out her PhD thesis at EMBL (Heidelberg). After her post-doctoral studies at UCSF (San Francisco), María started her own lab at the Institute of Cancer Research in London, where she identified mammary stem/progenitor cells in the human breast. At CIC bioGUNE, her team is characterising normal and cancer stem cells and examining their role in resistance to endocrine therapy. She is on the committee of the European Network for Breast Development and Cancer and has worked with breast cancer charities, including Breakthrough (UK) and Acambi (Spain).

Short talk description: Breast cancer is the most frequently diagnosed cancer and the leading cause of death from cancer in women worldwide. Breast cancer is a heterogeneous disease. Cancer cells cannot function in isolation: tumours are populated by epithelial cells, stromal cells and immune cells, as well as small populations of normal stem cells and cancer stem cells (CSCs). In particular, the presence of CSCs proves a challenge to therapy and their identification and characterisation in breast cancer has opened new possibilities for anti-cancer therapies. CSCs are implicated in tumour initiation and resistance to current treatments, including endocrine therapy. We have previously shown that increase of CSC content during development of resistance to tamoxifen in breast cancer is driven by multiple signals, including Sox2-dependent activation of Wnt signalling. These findings highlight the relevance of Sox family transcription factors as potential therapeutic targets in breast cancer.

Short cv: Prof Patricia Rousselle is research director at the National Centre for Scientific Research (CNRS) in France and her research is focused on understanding cell / extracellular matrix interactions in the context of skin biology. Developing both biochemical and cell biology approaches, her group is interested in analyzing molecular and cellular events occurring during wound repair, focusing on cell adhesion, migration processes governed by extracellular glycoproteins and proteoglycans. Her recent work has focused on the interactions of basal keratinocytes with extracellular matrix components during the re-epithelialisation phase of wound repair. She is currently past president of the French Society for Matrix Biology, and she is a member of the board of Directors of the European Tissue Repair Society (ETRS). Patricia Rousselle obtained a doctorate in Pharmacy in 1988 and pursued a four-year residency in clinical biology at Lyon University-Hospitals, where she also gained an MSc in Molecular and Cellular Biology in 1990. She started a doctoral training at the Shriners Hospital for Crippled Children in Portland, USA, where she discovered the important dermal epidermal junction protein laminin 332. She thereafter worked at the Cutaneous Biology Research Center in Boston, USA, before she joined the French CNRS in 1995 to set up her own group. Since then, she conducts her research team independently through research funding, partnerships with industry and scientific collaborations.

Short talk description: The formation of complex organs, the organization of cells within them, and the expression of tissue-specific genes require a combination of spatial instructions from the extracellular matrix and intercellular adhesions, together with temporal signals from soluble factors such as growth factors), hormones and cytokines. The precise integration of the resulting intracellular signaling networks drives the principal cellular processes of suppression of apoptosis, cell cycle, migration and differentiation, which are all key to the morphogenesis and function of tissues.

Θα θέλαμε να σας προσκαλέσουμε να συμμετάσχετε στη διαδικτυακή συνάντηση με τίτλο: «Η καινοτόμος δύναμη της Χημείας στην αντιμετώπιση των προκλήσεων του μέλλοντος», η οποία προγραμματίζεται να πραγματοποιηθεί την Τρίτη 21 Δεκεμβρίου και ώρα 18:00.

Η εκδήλωση εντάσσεται στο πλαίσιο των «Συναντήσεων Επιστήμης» που διοργανώνεται από το Ίδρυμα Ευγενίδου (www.eef.edu.gr) για τις Θετικές Επιστήμες (https://www.eef.edu.gr/el/to-idryma/online-draseis/synantiseis-epistimis/.και πλαισιώνεται από κορυφαίους επιστήμονες και μέλη της ακαδημαϊκής κοινότητας, ανάμεσα στους οποίους και δύο έγκριτα μέλη της Σχολής Θετικών Επιστημών του Πανεπιστημίου Πατρών, οι κ.κ. Συγκεκριμένα:
1.    ο κ. Νίκος Καραμάνος θα συντονίσει τη συνάντηση και θα εισηγηθεί σχετικά με τον «Κομβικό ρόλο της Χημείας στις μελλοντικές προκλήσεις της ανθρωπότητας»,
2.    ο κ. Χρήστος Κορδούλης θα επικεντρωθεί στην ομιλία του για τη «Χημεία ως πυλώνα βιώσιμης ανάπτυξης», ενώ
3.    ο κ. Ιωάννης Παπαευσταθίου θα μιλήσει για τη «Χημεία στην αντιμετώπιση των προκλήσεων του νερού».


Το θέμα της συνάντησης έχει σημαντικό διαθεματικό ενδιαφέρον μιας και η Χημεία αποτελεί τη βάση της ζωής και της φύσης. Είναι η επιστήμη που διασφαλίζει ότι έχουμε φάρμακα για την πρόληψη και τη θεραπεία των ασθενειών, επαρκή τρόφιμα τα οποία είναι ασφαλή,  προϊόντα προσωπικής φροντίδας και υγιεινής και τόσα άλλα.

 Όλα μέρη του κόσμου θα βιώσουν τις μεγάλες τάσεις που δημιουργούνται από τις προκλήσεις του μέλλοντος, λόγω της αύξησης του πληθυσμού, της κατανάλωσης των πόρων, και των ασθενειών. Το ερώτημα πως η Χημεία θα αντιμετωπίσει τις μελλοντικές επιπτώσεις είναι επίκαιρο και επιτακτικό. Στην επιστημονική αυτή συνάντηση θα συζητηθούν σημαντικές προκλήσεις για το μέλλον στις οποίες η Χημεία μπορεί να διαδραματίσει κομβικό ρόλο.

Short cv: Martin Götte is  a  Professor  at  the Department  of  Gynecology and  Obstetrics of the  University of Münster, Germany.  He obtained his diploma (Biology) in 1994 at the University of Münster, and his Ph.D.  (Biochemistry) in 1997 from the University of Göttingen and the Max-Planck-Institute for Biophysical Chemistry. After postdoctoral   research at the Department of Cell Biology of Harvard Medical School, Boston, USA, he obtained a tenured position as head of the research laboratory of the Department of Gynecology and Obstetrics of Münster University Hospital. His main areas of research are the role of syndecans, decorin, and glycosaminoglycan biosynthetic enzymes in cancer and inflammation. 

Short talk description: The malignant behavior of cancer cells is determined not only by cell-autonomous factors but also by a permissive microenvironment. The extracellular matrix (ECM) plays a major role in either supporting or inhibiting tumor cell behavior. Epigenetic mechanisms and a dysregulated endocrine milieu favor altered ECM synthesis. Aberrant production and function of cell surface proteoglycans conveys signals to tumor which promote metastatic behavior, unlimited tumor growth, and cancer stem cell function linked to therapeutic resistance and relapse. Proteoglycans mediate matrix-dependent signaling events via growth factors and chemokines, modulating angiogenesis and immune cell function in the tumor microenvironment. Moreover, they regulate the function of immune cells, shifting their function from an anti-cancerous to a permissive state. Focusing on the function of the cell surface proteoglycan family of syndecans, this lecture will provide basic knowledge on the function of these proteoglycans in signal transduction, matrix adhesion and degradation, and in regulating inflammatory processes and angiogenesis in the context of tumor progression.

Σύντομο βιογραφικό του ομιλητή.

Dr. Andrea Carfi holds a Master of Science in Physics from University of Canterbury in UK, Laura degree (equivalent to Master of Science in Chemistry) from Pavia University, Italy, and a PhD in Biophysics from the Université Joseph Fourier in Grenoble, France. He was also trained as a postdoctoral fellow in the laboratories of Prof. Stephen Harrison and Prof. Don Wiley at Boston Children’s Hospital/Harvard. Dr. Carfi has more than 18 years of experience in drug discovery and vaccine development at Moderna, GSK, Novartis Vaccines and IRBM/Merck. Over the years he has covered roles of increasing responsibility with a focus on structural biology, antigen design, small molecule antivirals discovery and vaccines development. In particular, during the seven years at GSK and Novartis Vaccines, he led the US based Protein Biochemistry team and was responsible for the design, selection, characterization and early development of novel vaccines against infectious disease targets, including HIV, CMV and RSV. Some of these vaccine candidates are now in early and late stage clinical trials. He was also heading the Novartis Vaccines Antigen Design platform across the research sites of Cambridge, US and Siena, Italy. Dr. Carfi joined Moderna in 2017 as Sr. Director of Antigen Design and Selection and Project Leader for the CMV vaccine program now in a Phase 1 clinical trial. Dr. Carfi has become the Head of Research for Infectious Disease since January 2019 and he is currently leading Research for all Moderna Infectious Disease targets.

Abstract: Artificial Intelligence (AI) has had a profound impact on areas such as image and speech recognition; however, comparable advances in drug discovery are still rare. This contribution will explore some of the reasons why this is the case, starting firstly by embedding the application of AI in drug discovery into its historical context. Next, we will explore which types of improvements – namely to speed, cost, or quality of decisions – are able to have the most profound impact on the success of drug discovery projects, and we will see that it is quality of decisions in clinical phases that matters most for project success. However, while computational algorithms are tremendously powerful today, much of the data we have available in drug discovery projects comes from the preclinical, proxy domain – which possesses only limited utility to make predictions for clinical stages. Hence, while numerically we are at least to an extent becoming better at modelling proxy endpoints, this leads to a ‘models of models’ situation, which only translates to a very limited extent to the safety and efficacy of compounds which will reach patients in a clinical setting. Even more fundamentally, there are intrinsic aspects of chemical and biological data which make labelling data much more difficult than in the image- or speech-domain, such as its conditionality (e.g. the dependence of effects on dose), which pose conceptual problems to being able to apply computational algorithms in the drug discovery domain. The contribution will conclude with aspects of model validation and perceptions of AI in society which pose additional problems for their unbiased evaluation and development, and point out developments which are needed for applications of AI in drug discovery to contribute to the clinical safety and efficacy of future medicines.

Short CV:
Dr Andreas Bender is Director for Digital Life Sciences at Nuvisan in Berlin, as well as a Reader for Molecular Informatics with the Centre for Molecular Science Informatics at the Department of Chemistry of the University of Cambridge. In his work, Andreas is involved with the integration and analysis of chemical and biological data from different sources, such as structural and bioactivity data, gene expression readouts, cellular imaging data, pathway information, etc. The computational analysis of this information is in his research then aimed at understanding phenotypic compound action – such as cellular readouts and organism-level effects – on a mechanistic level, predicting molecular properties related to both compound effiacy and toxicity, as well as e.g. compound repurposing. He received his PhD from the University of Cambridge and worked in the Lead Discovery Informatics group at Novartis in Cambridge/MA as well as at Leiden University in the Netherlands and at AstraZeneca before his current post.
